6-BA拮抗脱落酸缓解渗透胁迫对种子萌发的抑制

摘 要:

细胞分裂素促进细胞分裂、芽的分化, 拮抗脱落酸抑制的种子萌发, 而细胞分裂素合成基因Ipt8在种子萌发过程中发挥重要的作用。本文分别用120 mmol·L-1 NaCl和240 mmol·L-1甘露醇模拟盐和干旱胁迫处理拟南芥种子, 探讨6-BA拮抗ABA对其抑制种子萌发和萌发后生长的影响。结果表明, 细胞分裂素合成相关突变体ipt6-1ipt6-2ipt8-1ipt8-2的种子萌发和生长可被NaCl和甘露醇显著抑制; 而ABA合成相关突变体aba2-1对相同浓度NaCl和甘露醇的处理表现相对不敏感。进一步研究发现添加外源6-BA可恢复ipt6-1ipt6-2ipt8-1ipt8-2的相关敏感表型, 并且随6-BA浓度的增加, 恢复效果也愈趋明显。

Alleviation of 6-BA Antagonizing ABA on the Inhibition of Seed Germination by Osmotic Stress

Abstract:

Cytokinins promote cell division, differentiation of buds, antagonist abscisic acid inhibition of seed germination. The cytokinin biosynthesis gene Ipt8 plays an important role in the process of seed germination. In this paper, we studied the alleviation of 6-BA antagonizing ABA on the inhibition of seed germination in Arobidopsis thaliana by osmotic stress. And the Arobidopsis seeds were treated with 120 mmol·L-1 NaCl and 240 mmol·L-1 manntiol to simulate the salt and drought stress. The results showed that the seed germination and growth of cytokinin biosynthesis mutants ipt6-1, ipt6-2, ipt8-1, and ipt8-2 were significantly inhibited by NaCl and mannitol. But the ABA biosynthesis the mutant aba2 was insensitived to same concentrations of NaCl and manntiol. Further results showed that the sensitived ecotype of ipt6-1, ipt6-2, ipt8-1, ipt8-2 could be recovered by adding exogenous cytokinin (6-BA), otherwise the effects were more obvious with the increasing concentrations of 6-BA.
